O jogo irá ter 48 canções dos 5 primeiros jogos da série: Guitar Hero, Guitar Hero II, Guitar Hero Encore: Rocks the 80s, Guitar Hero III: Legends of Rock e Guitar Hero: Aerosmith. Todas as canções irão ter suporte para bateria e microfone, além da guitarra e do baixo. O jogo está previsto para ser lançado no dia 16 de junho de 2009 para PlayStation 2, PlayStation 3, Xbox 360 e Wii.
